吉林省粳稻种质萌发期耐碱性和耐盐性综合评价

Comprehensive Evaluation of Rice(Oryza sativa japonica) Germplasm for Alkaline/Saline Tolerance at Germination Stage from Jilin Province,China

  • 摘要: 收集具有代表性的吉林省不同熟期的粳稻种质资源共60份,采用培养皿发芽方法,在盐胁迫(120 mM NaCl,pH=6.98)和碱胁迫(30 mM Na2CO3,pH=11.05)条件下培养,并测定发芽势、发芽率、芽长、根长和根数等生长指标,利用隶属度对水稻种质萌发期耐盐碱性进行综合评价,为未来盐碱地水稻直播及抗逆育种提供抗逆种质资源。结果表明,水稻萌发期耐盐性和耐碱性呈极显著正相关(r=0.728**),但品种间存在差异。相对芽长和相对根长分别与抗盐隶属度(r芽长=0.862**,r根长=0.781**)和抗碱隶属度(r芽长=0.757**,r根长=0.812**)呈极显著正相关,可作为水稻萌发期耐盐碱性快速筛选的重要指标。利用抗逆隶属度指标将耐盐性和耐碱性划分成高抗(HT)、中抗(MT)、低抗(T)、敏感(S)、中度敏感(MS)和高度敏感(HS)共6个等级,初步筛选出耐盐且耐碱的品种主要有长白21号(HT/HT)、通禾835(HT/HT)、吉玉粳(HT/HT)、长白10号(HT/MT)、吉粘6号(MT/HT)、吉粘10号(MT/MT)、吉粳507(MT/MT)、吉粳509(MT/MT)、吉粳806(MT/MT)和东稻4号(MT/MT);耐盐不耐碱的品种有通禾99(HT/S)、通禾837(MT/S);耐碱不耐盐的品种有吉粳105(MT/S)、吉农大828(MT/S);不耐盐也不耐碱的品种有九稻51号(HS/HS)、吉农大888(HS/HS)和通禾834(HS/MS)等。同时发现,与晚熟或中晚熟品种相比,吉林省中早熟或中熟水稻品种表现出更好的耐盐碱性特征,这对今后水稻耐盐碱育种亲本资源选择具有重要的参考价值。

     

    Abstract: In order to provide excellent germplasm resources for direct seeding and breeding for saline and alkaline stress tolerance,60 representative local japonica rice cultivars from Jilin province,China,were evaluated for their tolerance to saline and alkaline stresses at seed germination stage based on the membership degree. The saline and alkaline stresses were simulated by 120 mM NaCl (pH=6.98) and 30 mM Na2CO3 (pH=11.05) solutions respectively,and rice seeds were germinated on filter papers soaked with different stress solutions in a petri-dish (9 cm diameter). Five indices of seed germination including germination potential,germination rate,plumule length,root length and root number were measured under stressed or unstressed control conditions. The results revealed that the correlation of resistance to saline and alkaline reached highly significant level (r=0.728**),though there are varietal differences. Relative plumule length,relative root length had good correlations with alkaline tolerance (rBL=0.757**,rRL=0.812**) and saline tolerance (rBL=0.862**,rRL=0.781**),representing a useful set of simple and fast monitoring parameters for assessing saline/alkaline tolerance levels for individual rice varieties. Based on the membership degree,the varieties were divided into 6 grades of saline or alkaline tolerance:highly tolerant (HT),moderately tolerant (MT),tolerant (T),susceptible (S),moderately susceptible (MS),highly susceptible (HS). We identified varieties Changbai 21 (HT/HT),Tonghe 835 (HT/HT),Ji Yujing (HT/HT),Changbai1 0(HT/MT),Jinian 6 (MT/HT),Jinian 10 (MT/MT),Jijing 507 (MT/MT),Jijing 509 (MT/MT),Jijing 806 (MT/MT) and Dongdao 4 (MT/MT) as tolerant to both the saline and alkaline; Tonghe 99 (HT/S)and Tonghe 837(MT/S) as tolerant to saline but susceptible to alkaline; Jijing 105 (MT/S) and Ji Nongda 828 (MT/S) as tolerant to alkaline but susceptible to saline; Jiudao 51 (HS/HS),Ji Nongda 888 (HS/HS)and Tonghe 834(HS/MS) as susceptible to both the saline and alkaline stresses. Furthermore,compared with the mid-late maturing or late maturing rice varieties,the early-medium maturing or medium maturing rice varieties in Jilin province showed better saline-alkaline tolerant performance. This finding may provide important reference for selecting parent sources in the saline-alkaline tolerant rice breeding programs.
